**Alvocidib + cytarabine + mitoxantrone** is a timed-sequential multi-agent chemotherapy regimen studied for the treatment of relapsed/refractory and high-risk newly diagnosed acute myeloid leukemia (AML), including MCL-1-dependent subtypes. **Alvocidib** is a potent small molecule inhibitor of cyclin-dependent kinase 9 (CDK9), resulting in suppression of genes critical for leukemia cell survival (notably MCL-1)[7][3]. **Cytarabine** is a nucleoside analog that inhibits DNA synthesis, and **mitoxantrone** is an anthracenedione that intercalates into DNA and inhibits topoisomerase II, leading to DNA damage and apoptosis[9][1][3]. The ACM regimen has demonstrated significant clinical activity and tolerability in clinical trials among AML patients with poor prognostic factors, as well as superior cancer reduction compared to standard induction therapies in certain studies[3][1][5]. This regimen is under clinical investigation, with developer support from Tolero Pharmaceuticals (now Sumitomo Dainippon Pharma), and has no distinct brand/trade name beyond its constituent drugs.
