Overview

AM001 is a topical cream formulation of apremilast, a small molecule phosphodiesterase-4 (PDE4) inhibitor, developed by AmDerma for the treatment of moderate to severe plaque psoriasis. PDE4 is an enzyme responsible for the degradation of cyclic adenosine monophosphate (cAMP) in inflammatory cells; its inhibition leads to increased intracellular cAMP levels, which subsequently downregulates the production of pro-inflammatory cytokines (such as TNF-alpha, IL-17, and IL-23) and upregulates anti-inflammatory mediators. By delivering apremilast topically, AM001 aims to provide localized efficacy at the site of psoriatic plaques while potentially avoiding the systemic gastrointestinal side effects, such as nausea and diarrhea, frequently associated with oral apremilast therapy.
