Overview

AMA1(D12)-RON2L-TM mRNA-LNP vaccine is a preclinical mRNA-lipid nanoparticle (LNP) vaccine candidate designed to target Plasmodium falciparum malaria. Developed by researchers at Johns Hopkins University, the National Institutes of Health (NIH), and the University of Victoria, the vaccine utilizes a structure-guided design to overcome the high antigenic polymorphism of Apical Membrane Antigen 1 (AMA1). The construct consists of AMA1 domains 1 and 2 (D12), with domain 3 eliminated to focus the immune response on neutralizing epitopes. It is fused to the RON2L peptide (a 49-amino acid region of Rhoptry Neck Protein 2, the parasite-derived binding partner of AMA1) to mimic the functional receptor-ligand complex and enhance the production of cross-neutralizing, strain-transcending antibodies. Additionally, the construct incorporates a transmembrane region (TM) for membrane anchoring, which enhances antibody titers and directs the immune response to the apical end of AMA1.
