Overview

Amatuximab (MORAb-009) is a chimeric monoclonal IgG1/k antibody that targets mesothelin, a cell-surface glycoprotein overexpressed in several cancers, including mesothelioma, pancreatic ductal adenocarcinoma, ovarian cancer, and non-small cell lung cancer. Developed by Morphotek (later acquired by Eisai) in collaboration with the National Cancer Institute (NCI), amatuximab works by binding to mesothelin and inhibiting its interaction with CA125 (MUC16), a protein involved in cell adhesion and tumor progression. It also potentially induces antibody-dependent cellular cytotoxicity (ADCC). In clinical research, a radiolabeled version (111In-amatuximab) was developed to study the biodistribution and safety of the antibody in patients with mesothelin-expressing tumors. Although it reached Phase 2 clinical trials, development was ultimately discontinued by Eisai after failing to meet primary efficacy endpoints in mesothelioma.
