This is a combination therapy consisting of amatuximab (a chimeric IgG1 monoclonal antibody targeting mesothelin), cisplatin (a platinum-based chemotherapy agent), and pemetrexed (a folate analog). The combination targets mesothelin-expressing cancers, particularly malignant pleural mesothelioma. Amatuximab works by binding to mesothelin, blocking its interaction with CA125/MUC16, and triggering antibody-dependent cellular cytotoxicity (ADCC). This prevents cell adhesion, migration patterns, and metastasis. Pemetrexed inhibits multiple enzymes involved in folate metabolism and DNA synthesis, while cisplatin acts as a platinum-based chemotherapeutic agent.
