Overview

**Amatuximab + gemcitabine** is a combination regimen under clinical investigation for solid tumors expressing mesothelin, most notably pancreatic cancer and mesothelioma. Amatuximab is a chimeric monoclonal antibody that targets mesothelin, a cell surface glycoprotein highly expressed on certain cancers. Gemcitabine is a nucleoside analog (small molecule chemotherapeutic) that inhibits DNA synthesis. Amatuximab mediates antitumor activity primarily via antibody-dependent cellular cytotoxicity (ADCC) by binding to mesothelin; preclinical and clinical studies show enhanced antitumor effect and synergistic killing when combined with gemcitabine. The combination is being tested primarily for its potential to suppress metastasis and improve outcomes in patients with mesothelin-expressing tumors. There have been phase II clinical trials for pancreatic cancer and mesothelioma, but with mixed or modest efficacy signals[2][3][4][5][6][7].
