Overview

**Amcasertib + doxorubicin** is a combination regimen under investigation for cancer therapy, consisting of amcasertib, a small molecule inhibitor of cancer stemness kinases (notably NANOG and PDGFRα), and doxorubicin, an anthracycline chemotherapeutic agent that intercalates DNA, inhibits topoisomerase II, and induces apoptosis. Amcasertib is given orally and targets pathways central to cancer cell stemness and drug resistance[5][6][9]. Doxorubicin is administered intravenously and is widely used for a variety of cancers; in the context of this combination, the aim is to enhance anti-tumor efficacy by targeting both cancer stem cells and bulk tumor cells. The combination is being explored in solid tumors, including ovarian, liver, and cholangiocarcinoma, as both agents show synergy and potential to overcome resistance[6][9].
