**Amcasertib + nivolumab** is an investigational combination of two anti-cancer agents with complementary mechanisms of action. **Amcasertib** (also known as BBI-503) is an orally active, first-in-class small molecule inhibitor that targets multiple cancer stemness kinases, notably inhibiting the Nanog pathway and several serine-threonine kinases, thereby suppressing stem-cell-like features and inducing apoptosis in cancer stem cells[2][3][4][5][6]. It has demonstrated cytotoxicity, reduction of epithelial-mesenchymal transition (EMT), and inhibition of cell invasion and migration in preclinical studies. **Nivolumab** (marketed as Opdivo) is a human monoclonal antibody that inhibits Programmed cell death protein 1 (PD-1), thereby boosting anti-tumor immunity. The rationale for combining these drugs is to target both the stemness potential of resistant cancer cells (amcasertib) and activate adaptive immune response against tumor cells (nivolumab). This combination is being studied for various advanced cancers including breast, ovarian, and other solid tumors[2][3][5].
