Amcenestrant + palbociclib is a combination therapy investigated for the first-line treatment of estrogen receptor-positive, human epidermal growth factor receptor 2-negative (ER+/HER2-) advanced or metastatic breast cancer. Amcenestrant is an investigational oral selective estrogen receptor degrader (SERD) that antagonizes and degrades the estrogen receptor, thereby inhibiting ER signaling pathways. Palbociclib is a small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), which blocks cell cycle progression from G1 to S phase in cancer cells. The AMEERA-5 phase III trial compared this combination to letrozole plus palbociclib but was discontinued at interim analysis due to futility; no new safety signals were identified[1][2][3][8]. The development program for amcenestrant has since been discontinued by Sanofi[2].
