Overview

AMD3100, also known as plerixafor or Mozobil, is a small-molecule antagonist of the chemokine receptor CXCR4. It blocks the interaction between CXCR4 and its ligand CXCL12, thereby inhibiting chemotaxis and mobilizing hematopoietic stem/progenitor cells from the bone marrow into peripheral blood. It is FDA-approved for use in patients with non-Hodgkin's lymphoma and multiple myeloma to facilitate stem cell transplantation[5][7]. In preclinical studies, topical application of AMD3100 has been shown to promote wound healing by increasing cytokine production and enhancing fibroblast migration and proliferation[1]. Recombinant human PDGF-BB (rhPDGF-BB) is a biologically active dimeric protein that stimulates cell proliferation, migration, and survival primarily through binding to PDGF receptors alpha and beta. It acts as a potent mitogen for connective tissue cells such as dermal fibroblasts, smooth muscle cells, glial cells, some epithelial/endothelial cells, and plays roles in wound healing by promoting granulation tissue formation[3][8]. The combination of AMD3100 + rhPDGF-BB has been studied experimentally for synergistic effects on wound healing—AMD3100 enhances recruitment of progenitor/fibroblast populations while rhPDGF-BB directly stimulates their proliferation.
