Overview
Amenamevir (ASP2151) is a non-nucleoside small molecule antiviral drug developed by Maruho Co., Ltd. It acts as a helicase-primase inhibitor, targeting the viral helicase-primase enzyme complex, which is essential for the replication of herpes viral genomic DNA. Amenamevir has demonstrated antiviral activity against herpes simplex virus types 1 and 2 (HSV-1, HSV-2) and varicella-zoster virus (VZV). It is marketed under the brand name Amenalief in Japan for the treatment of herpes zoster and herpes simplex virus infections. Ciclosporin (cyclosporine) is a calcineurin inhibitor used as an immunosuppressant, particularly in patients undergoing organ and tissue transplantation. The combination of ASP2151 and ciclosporin is specifically associated with a Phase 1 drug-drug interaction (DDI) study conducted by Maruho Europe Limited to investigate the effect of repeated oral doses of ciclosporin (a CYP3A inhibitor) on the single-dose pharmacokinetics of amenamevir (a CYP3A4 substrate and inducer) in healthy volunteers.
