Overview
AMG 330 + pembrolizumab is an investigational combination therapy for adult subjects with relapsed or refractory acute myeloid leukemia (AML). AMG 330 is a bispecific T-cell engager (BiTE) antibody targeting CD33 on AML cells and CD3 on T cells, facilitating immune-mediated cytotoxicity of CD33+ AML blasts. Pembrolizumab is a humanized IgG4 monoclonal antibody that blocks programmed cell death protein 1 (PD-1), inhibiting its interaction with PD-L1 and PD-L2, and serving as an immune checkpoint inhibitor. The rationale for combining these agents is based on evidence that AMG 330 induces up-regulation of PD-L1 on AML cells and PD-1 on T cells, potentially limiting anti-leukemic activity. Blocking the PD-1/PD-L1 axis with pembrolizumab is hypothesized to enhance and prolong the effectiveness of AMG 330 by overcoming acquired resistance and augmenting T cell activation[1][3][4].
