Overview

AMG 655 + bortezomib is a combination therapy investigated primarily in early-phase clinical trials for lymphoma and other cancers. AMG 655, also known as conatumumab, is a fully human monoclonal antibody that acts as an agonist of the death receptor 5 (DR5 or TRAIL-R2), promoting apoptosis in tumor cells through activation of the extrinsic apoptotic pathway. Bortezomib is a small-molecule proteasome inhibitor that induces apoptosis by preventing the degradation of pro-apoptotic factors and sensitizing tumor cells to death receptor signaling. Combining AMG 655 with bortezomib is intended to enhance anti-tumor efficacy, particularly by overcoming resistance to death receptor-mediated apoptosis observed in some cancer cells[1][2][5][7].
