Overview

Amifostine + fluorouracil is a combination regimen used in oncology, particularly for patients undergoing chemotherapy for cancers such as metastatic colorectal carcinoma. Amifostine is a cytoprotective agent that acts as a selective free radical scavenger, protecting normal tissues from the toxic effects of chemotherapy and radiation by detoxifying reactive metabolites and scavenging free radicals. Fluorouracil (5-fluorouracil or 5-FU) is an antimetabolite chemotherapeutic agent that inhibits thymidylate synthase, thereby disrupting DNA synthesis and leading to cell death in rapidly dividing cancer cells. The combination aims to reduce the gastrointestinal and mucosal toxicities associated with high-dose fluorouracil therapy without compromising its anticancer efficacy[1][3][4].
