Overview

Amifostine + vinorelbine is a combination regimen of two pharmaceutical agents used primarily in oncology. Amifostine is a cytoprotective adjuvant that acts as a prodrug, which is dephosphorylated in tissues to an active thiol metabolite that scavenges free radicals and protects normal tissues from the toxic effects of chemotherapy and radiation, particularly nephrotoxicity and myelosuppression. Vinorelbine is a semi-synthetic vinca alkaloid antineoplastic agent that inhibits microtubule assembly by binding to tubulin, thereby disrupting mitosis and leading to cell cycle arrest at metaphase. This combination has been studied mainly for its potential to reduce toxicity (especially renal toxicity) associated with chemotherapeutic regimens in non-small cell lung cancer (NSCLC), often as part of multi-drug protocols or alongside radiotherapy[1][3][5]. Amifostine does not have direct antitumor activity but serves as a protective adjunct; vinorelbine provides the cytotoxic effect.
