Overview

Amikacin + colistin is a combination of two antibiotics—amikacin, an aminoglycoside, and colistin, a polymyxin—used primarily for the treatment of severe infections caused by multidrug-resistant Gram-negative bacteria. This combination has been studied for its synergistic effects against pathogens such as Pseudomonas aeruginosa and carbapenem-resistant Klebsiella pneumoniae (CRKP), especially in biofilm-associated and device-related infections. Amikacin acts by binding to the 30S ribosomal subunit, inhibiting protein synthesis in bacteria, while colistin disrupts the bacterial outer membrane by interacting with lipopolysaccharides. The dual mechanisms enhance bactericidal activity and may help overcome resistance seen with monotherapy[1][2][4][6].
