Overview

A combination of aminoglutethimide and danazol that has been studied for the treatment of metastatic breast cancer. This combination demonstrates biochemical antagonism, with aminoglutethimide inhibiting adrenal steroid synthesis while danazol functions as a synthetic steroid and pituitary gonadotropin inhibitor. Clinical studies have shown poor efficacy of this combination compared to aminoglutethimide alone in advanced breast cancer patients, likely due to danazol opposing the suppressive effects of aminoglutethimide on biologically-active estradiol.
