A combination therapy consisting of aminoglutethimide, an adrenocortical steroid synthesis inhibitor, and hydrocortisone, a glucocorticoid. This combination works by inhibiting the production of adrenal steroids while providing necessary glucocorticoid replacement. Aminoglutethimide blocks enzymes including Cholesterol side-chain cleavage enzyme (CYP11A1) and Aromatase (CYP19A1), thereby inhibiting the conversion of cholesterol into steroid hormones and blocking the production of androgens, estrogens, and glucocorticoids. Hydrocortisone is added to replace depleted glucocorticoids and prevent adrenal insufficiency.
