Overview

Aminophylline + sunitinib is a combination of two small molecule drugs with distinct mechanisms of action. Aminophylline is a bronchodilator composed of theophylline and ethylenediamine, primarily used to treat bronchospasm in conditions such as asthma, emphysema, and chronic bronchitis. Its mechanism involves inhibition of phosphodiesterase (PDE) enzymes, antagonism at adenosine receptors (A1 and A3), and activation of histone deacetylase 2, leading to bronchodilation and anti-inflammatory effects[1]. Sunitinib is an oral multikinase inhibitor approved for the treatment of cancers such as gastrointestinal stromal tumor (GIST) and advanced renal cell carcinoma. It targets multiple receptor tyrosine kinases including vascular endothelial growth factor receptors (VEGFRs), platelet-derived growth factor receptors (PDGFRs), stem cell factor receptor (c-KIT), FMS-like tyrosine kinase 3 (FLT3), colony-stimulating factor 1 receptor (CSF1R), and glial cell line-derived neurotrophic factor receptor. This results in inhibition of tumor angiogenesis and proliferation[4]. There is no evidence that this specific combination has been developed or approved for any indication.
