Overview

amiodarone + CoSeal is an experimental drug-device combination consisting of the Class III antiarrhythmic agent amiodarone hydrochloride incorporated into the CoSeal Surgical Sealant, a synthetic polyethylene glycol (PEG) hydrogel. Developed by researchers at the University of Louisville, this formulation is designed for intra-operative application to the epicardial surface of the atria and pulmonary veins during cardiothoracic surgeries, such as lung transplantation. The hydrogel serves as a localized, sustained-release delivery system, aiming to prevent postoperative atrial fibrillation (POAF) by maintaining high local concentrations of amiodarone while reducing the systemic toxicity typically associated with oral or intravenous administration. The amiodarone component acts by blocking multiple ion channels (potassium, sodium, and calcium) and exerting non-competitive anti-adrenergic effects, thereby stabilizing cardiac rhythm.
