Overview

Amiselimod + corticosteroid refers to the co-administration of amiselimod, a selective sphingosine 1-phosphate receptor-1 (S1P1) modulator, with a corticosteroid (most commonly prednisolone or a similar glucocorticoid). Amiselimod, developed as MT-1303, acts primarily as an S1P1 (and to a lesser extent S1P5) functional antagonist; it is an oral small molecule immunomodulator, reducing circulating lymphocytes by trapping them in secondary lymphoid organs. This reduces immune-mediated inflammation, with a favorable cardiac safety profile compared to fingolimod. Corticosteroids are anti-inflammatory and immunosuppressive agents acting mainly via glucocorticoid receptors, leading to gene transcription changes that suppress pro-inflammatory signals and promote anti-inflammatory processes. This drug combination is evaluated for the treatment of autoimmune diseases such as systemic lupus erythematosus (SLE) and ulcerative colitis, in which patients often receive both drugs together to achieve disease control.
