Overview

Amisulpride + risperidone is a combination of two atypical antipsychotic drugs used primarily in the management of schizophrenia, particularly in patients who are partially or not fully responsive to monotherapy. Amisulpride acts as a selective antagonist at dopamine receptor D2 and dopamine receptor D3, while risperidone antagonizes both dopamine receptor D2 and serotonin 5-HT2A receptor, with additional affinity for adrenergic receptor alpha 1 and histamine H1 receptor. The combination may be considered as an augmentation strategy in treatment-resistant schizophrenia to improve symptom control. However, co-administration increases the risk of serious cardiac arrhythmias due to potential additive effects on QT prolongation[1][2][3]. Both agents are small molecule pharmaceuticals developed by different companies (amisulpride by Sanofi-Aventis; risperidone by Janssen), but their combination is not marketed as a fixed-dose product.
