Overview

Amitriptyline + haloperidol is a combination of two pharmaceutical drugs, amitriptyline and haloperidol. Amitriptyline is a tricyclic antidepressant that inhibits the reuptake of norepinephrine and serotonin, increasing their synaptic concentrations in the central nervous system. It also has anticholinergic and sedative properties. Haloperidol is a first-generation (typical) antipsychotic that primarily acts as an antagonist at dopamine D2 receptors, reducing dopaminergic neurotransmission in the brain. This combination has been studied for use in patients with combined depressive and psychotic syndromes, such as major depression with psychotic features or schizoaffective disorder (depressive type). Clinical trials have shown that this combination can lead to greater reductions in both depressive and psychotic symptoms compared to some atypical antipsychotics like risperidone[2][3][5]. However, additive central nervous system depression may occur when these drugs are used together[1].
