AmNurite P is an oral fixed-dose combination therapy developed by Health N U Therapeutics, consisting of the tricyclic antidepressant amitriptyline hydrochloride and the gabapentinoid pregabalin in a sustained-release (SR) formulation. This combination is primarily indicated for the management of neuropathic pain, such as diabetic peripheral neuropathy and postherpetic neuralgia. Amitriptyline acts by inhibiting the reuptake of serotonin and norepinephrine in the synaptic cleft, thereby enhancing descending inhibitory pain pathways. Pregabalin binds to the alpha-2-delta subunit of voltage-gated calcium channels in the central nervous system, reducing the release of excitatory neurotransmitters like glutamate and substance P. Together, these agents provide synergistic analgesic effects for chronic pain conditions, with the sustained-release formulation of pregabalin allowing for optimized dosing schedules.
