Amivantamab + FOLFIRI is a combination therapy consisting of amivantamab (a bispecific antibody targeting EGFR and MET) with FOLFIRI chemotherapy (folinic acid, fluorouracil, and irinotecan). This combination has shown promising rapid and durable antitumor activity in patients with RAS/BRAF wild-type metastatic colorectal cancer who have not previously received anti-EGFR therapy. In clinical trials, the combination demonstrated an overall response rate of 49%, median duration of response of 7.4 months, and median progression-free survival of 7.5 months. The safety profile is manageable and consistent with each individual component, without additive toxicity. Common treatment-emergent adverse events include neutropenia, rash, stomatitis, infusion-related reactions, and diarrhea.
