Drug intelligence / Profile preview
Amivantamab + folinic acid + fluorouracil + oxaliplatin is an investigational combination regimen consisting of the bispecific monoclonal antibody amivantamab (Rybrevant) and the standard FOLFOX chemotherapy regimen (folinic acid, fluorouracil, and oxaliplatin). Developed by Johnson & Johnson, this combination is being evaluated for the treatment of patients with RAS/BRAF wild-type metastatic colorectal cancer (mCRC). Amivantamab targets both the epidermal growth factor receptor (EGFR) and mesenchymal-epithelial transition (MET) receptor pathways, aiming to overcome resistance mechanisms associated with standard anti-EGFR therapies. The chemotherapy components provide cytotoxic activity: fluorouracil acts as an antimetabolite that inhibits thymidylate synthase (potentiated by folinic acid), while oxaliplatin is a platinum-based alkylating agent that induces DNA damage. This combination is currently being evaluated in Phase 3 clinical trials (such as the OrigAMI-2 study) as a first-line treatment for patients with unresectable or metastatic left-sided colorectal cancer.
