Drug intelligence / Profile preview
Rybrevant Faspro + pembrolizumab + carboplatin is an investigational combination regimen consisting of the subcutaneous bispecific antibody Rybrevant Faspro (amivantamab and hyaluronidase-lpuj), the PD-1 checkpoint inhibitor pembrolizumab (Keytruda), and the platinum-based chemotherapy carboplatin. Amivantamab is a fully human bispecific antibody targeting epidermal growth factor receptor (EGFR) and mesenchymal-epithelial transition (MET) receptor tyrosine kinases, co-formulated with recombinant human hyaluronidase PH20 to enable rapid subcutaneous administration. Pembrolizumab is a monoclonal antibody that blocks the interaction between PD-1 and its ligands, enhancing T-cell-mediated anti-tumor immune responses. Carboplatin is a cytotoxic alkylating agent that causes DNA cross-linking, inhibiting DNA replication and transcription. This combination is being evaluated as a first-line treatment for patients with recurrent or metastatic head and neck squamous cell carcinoma (HNSCC), particularly those with human papillomavirus (HPV)-unrelated disease, in the Phase 1b/2 OrigAMI-4 and Phase 3 OrigAMI-5 clinical trials.
