Overview
This combination therapy consists of amivantamab, a bispecific antibody targeting EGFR and MET; lazertinib, a third-generation EGFR tyrosine kinase inhibitor; and pemetrexed, a folate antimetabolite chemotherapy. It is being investigated as a first-line treatment for patients with recurrent or metastatic non-small cell lung cancer (NSCLC) harboring EGFR mutations, specifically exon 19 deletions or L858R point mutations. The regimen aims to provide comprehensive inhibition of the EGFR and MET pathways while utilizing cytotoxic chemotherapy to overcome or delay resistance. The Latin American Cooperative Oncology Group (LACOG) is the lead sponsor for the Phase 2 trial (LACOG-0121) evaluating this triplet regimen.
