Overview

Amivantamab + pembrolizumab is an experimental combination of two distinct anticancer drugs. **Amivantamab** is a bispecific monoclonal antibody that targets both the epidermal growth factor receptor (EGFR) and the mesenchymal-epithelial transition factor (MET) to block ligand binding, inhibit signaling pathways, and mark tumor cells for immune-mediated destruction. **Pembrolizumab** is a monoclonal antibody that blocks programmed cell death protein 1 (PD-1), serving as an immune checkpoint inhibitor to reinvigorate T-cell activity against tumor cells. Preclinical studies in squamous cell cancers (head and neck, lung) have shown this combination can remodel the tumor microenvironment, overcome resistance to PD-1 blockade (observed in pembrolizumab alone), enhance infiltration and activation of cytotoxic memory CD8 T cells, and suppress immunosuppressive metabolic adaptation via glycolytic pathway markers. This combination is currently under investigation for recurrent/metastatic head and neck squamous cell carcinoma and other solid tumors[1][2][3][7].
