Overview

This refers to a combination therapy used in clinical trials, where patients are randomized to receive either a tablet containing both amlodipine (a calcium channel blocker antihypertensive) and folic acid (a B vitamin involved in homocysteine metabolism), or a matching placebo. Amlodipine lowers blood pressure by inhibiting calcium influx into vascular smooth muscle and cardiac muscle, leading to vasodilation. Folic acid reduces plasma homocysteine levels, which is associated with reduced cardiovascular risk. The combination has been studied for its efficacy in lowering both blood pressure and total homocysteine among hypertensive patients with hyperhomocysteinemia, particularly those intolerant to ACE inhibitors[1][2][5][6]. Placebo serves as an inactive comparator in these studies.
