Overview

Amorphous calcium carbonate (ACC) is the least stable and most soluble polymorph of calcium carbonate. It is characterized by its amorphous molecular structure and nanometric particle size, which confer significantly higher solubility and bioavailability compared to crystalline forms of calcium carbonate. ACC rapidly dissolves in acidic environments to release bioavailable Ca2+ ions and basic carbonate anions that can be converted into bicarbonate for local pH buffering. This property underlies its use as a dietary supplement for improving bone, muscle, and heart strength; supporting bone health; reducing pain associated with bone disorders; and potentially modulating inflammation through local pH normalization. ACC has been investigated as a biomaterial for drug delivery systems, tissue engineering scaffolds, immunomodulation in oncology settings, tumor imaging enhancement (via CO2 generation), osteoporosis management (by reducing bone resorption), osteoarthritis treatment (by counteracting joint acidity), hypoparathyroidism therapy, and other metabolic or inflammatory diseases. The Israeli biotech company Amorphical has developed stabilized forms of ACC for clinical use in humans[1][2][4][5][6][8].
