amoxicillin + clavulanic acid (GSK)

Amoxicillin + clavulanic acid (marketed under the brand name Augmentin DuoForte) is a fixed-dose combination antibiotic containing amoxicillin (875 mg, as trihydrate) and clavulanic acid (125 mg, as potassium clavulanate). Amoxicillin is a broad-spectrum beta-lactam antibiotic that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs). Clavulanic acid is a beta-lactamase inhibitor that structurally mimics beta-lactam antibiotics and covalently binds to the active site of beta-lactamase enzymes, protecting amoxicillin from enzymatic degradation by resistant bacteria. Developed by GSK, Augmentin DuoForte is approved in Australia for the short-term treatment of various bacterial infections, including urinary tract, respiratory tract, and skin/skin structure infections. Additionally, it has been evaluated in a Phase 3 pilot trial (the TRAIN trial) sponsored by Townsville Hospital in Australia to assess the feasibility of treating mild acute uncomplicated appendicitis with antibiotics as an alternative to standard laparoscopic appendicectomy.
