Amphetamine-dextroamphetamine is a central nervous system stimulant composed of a combination of amphetamine and dextroamphetamine salts. It is currently being investigated in a Phase 4 randomized controlled trial led by Dr. Eva Szigethy at the University of Pittsburgh Medical Center (UPMC) for the treatment of cognitive impairment, specifically "brain fog," in patients with post-acute COVID-19 syndrome (Long-COVID). The drug's mechanism of action involves increasing the synaptic concentrations of the neurotransmitters dopamine and norepinephrine by acting as an agonist at the trace amine-associated receptor 1 (TAAR1) and interacting with the vesicular monoamine transporter 2 (VMAT2) to promote neurotransmitter release and inhibit reuptake. In the clinical trial context, it is administered at a 10mg daily dose and compared against a digital behavioral health intervention called RxWell to evaluate its efficacy in managing cognitive symptoms associated with Long-COVID.
