Amphetamine + SPD503 is a combination therapy consisting of a psychostimulant (amphetamine or its salts) and guanfacine hydrochloride extended-release (SPD503). This combination is used in the treatment of attention-deficit/hyperactivity disorder (ADHD), particularly in children and adolescents who have an inadequate response to stimulant monotherapy. Amphetamines act as central nervous system stimulants by increasing synaptic concentrations of norepinephrine and dopamine through promoting their release and inhibiting reuptake. Guanfacine is a selective alpha-2A adrenergic receptor agonist that modulates prefrontal cortical circuits involved in attention and impulse control. The combination has been shown to improve ADHD symptoms more than either agent alone, with no significant pharmacokinetic interactions between the two drugs[1][3][7].
