Amphetamine + methylphenidate is a hypothetical combination of two central nervous system (CNS) stimulants, amphetamine and methylphenidate. Both are widely used individually for the treatment of attention deficit hyperactivity disorder (ADHD) and narcolepsy. Amphetamines act primarily by increasing the release of norepinephrine and dopamine in the brain, mainly through interactions with trace amine-associated receptor 1 (TAAR1) and vesicular monoamine transporter 2 (VMAT2), leading to increased synaptic concentrations of these neurotransmitters[1][5]. Methylphenidate acts as a norepinephrine–dopamine reuptake inhibitor (NDRI), blocking the reuptake transporters for these neurotransmitters, thereby increasing their levels in synapses[8][6]. The combination would theoretically provide both mechanisms—enhanced release and inhibited reuptake—potentially resulting in additive or synergistic stimulant effects. However, there is no evidence that such a fixed-dose combination product is currently approved or marketed.
