Amphinex + bleomycin

Amphinex + bleomycin is an investigational combination therapy that utilizes the photosensitizer Amphinex (fimaporfin) together with the cytotoxic agent bleomycin in a technique called photochemical internalisation (PCI). In this approach, Amphinex is administered and then activated by light at the tumor site. This activation disrupts endosomal membranes within cancer cells and enhances the intracellular delivery of large therapeutic molecules such as bleomycin. Bleomycin itself is a glycopeptide antibiotic with antitumor activity that acts by inhibiting DNA synthesis and causing DNA strand breaks. The combination aims to increase the efficacy of chemotherapy while potentially reducing systemic toxicity. The primary indication under investigation has been local recurrence or advanced/metastatic cutaneous or subcutaneous malignancies, including head and neck cancers[2][3][5][7].
