Overview

Amprenavir + ritonavir is a combination of two antiretroviral drugs used in the treatment of HIV-1 infection. Both are protease inhibitors that act by binding to and inhibiting the HIV-1 protease enzyme, which is essential for processing viral polyprotein precursors into functional proteins required for infectious viral particles. Inhibition leads to the production of immature, non-infectious virus. Ritonavir is primarily used at low doses as a pharmacokinetic booster; it inhibits cytochrome P450 3A4 (CYP3A4), thereby increasing plasma concentrations and effectiveness of amprenavir. This combination is typically administered with other antiretrovirals as part of highly active antiretroviral therapy (HAART). Amprenavir was originally developed by Glaxo Wellcome (now GSK), while ritonavir was developed by AbbVie[1][3][7]. The use of this combination has largely been replaced in clinical practice by newer agents due to tolerability and pill burden concerns.
