Overview

Combination of **amprenavir** and **saquinavir**, both oral HIV-1 protease inhibitors, used together as a dual protease inhibitor regimen in HIV-1-infected individuals. This combination may be considered for salvage therapy in patients who have experienced failure with other antiretroviral therapies. Amprenavir and saquinavir both inhibit the HIV-1 protease enzyme, preventing viral polyprotein cleavage and resulting in the production of non-infectious immature viral particles. When combined, they are typically used with ritonavir as a pharmacokinetic booster to enhance exposure of both protease inhibitors. Clinical studies indicate that this combination, with ritonavir boosting, can lead to reductions in HIV-1 viral RNA and increases in CD4+ T-cell counts in treatment-experienced patients. Drug-drug interactions may occur, impacting plasma concentrations of each agent, so individualized dosing and pharmacokinetic monitoring are often required[1][3][2].
