Amrubicin + cisplatin is a chemotherapy regimen combining the synthetic anthracycline derivative amrubicin with the platinum-based agent cisplatin. This combination has been investigated primarily as a first-line treatment for extensive-disease small-cell lung cancer (ED-SCLC), where it demonstrated non-inferiority to the standard etoposide plus cisplatin (EP) regimen in randomized phase III trials. The mechanism of action involves DNA intercalation and topoisomerase II inhibition by amrubicin, and DNA cross-linking by cisplatin, leading to cell cycle arrest and apoptosis in rapidly dividing cancer cells[1][2][3]. The most common adverse events include neutropenia, leukopenia, bone marrow failure, pyrexia, and fatigue; these are generally manageable[1][2].
