Overview

Amrubicin + irinotecan is a combination chemotherapy regimen consisting of two small molecule cytotoxic agents used primarily in the treatment of lung cancers, including small-cell lung cancer (SCLC) and non-small cell lung cancer (NSCLC). Amrubicin is a synthetic anthracycline derivative that acts as a topoisomerase II inhibitor, while irinotecan is a camptothecin analog that inhibits topoisomerase I. The rationale for combining these agents lies in their complementary mechanisms of action targeting different DNA topoisomerases, potentially resulting in synergistic antitumor effects. This combination has been studied in both first-line and relapsed settings for extensive-disease SCLC and advanced NSCLC. Clinical trials have demonstrated notable response rates but also significant hematologic toxicities such as neutropenia and febrile neutropenia[1][2][5][8].
