Overview

Amrubicin + vinorelbine is a combination chemotherapy regimen consisting of amrubicin, a synthetic 9-aminoanthracycline derivative and topoisomerase II inhibitor, and vinorelbine, a semi-synthetic vinca alkaloid that disrupts microtubule dynamics. This combination has been studied primarily for the treatment of non-small cell lung cancer (NSCLC) as second-line therapy. The recommended dose for this combination is amrubicin 35 mg/m² and vinorelbine 15 mg/m². Amrubicin works by inhibiting topoisomerase II, preventing DNA replication and transcription in cancer cells. Vinorelbine binds to β-tubulin at the Vinca-binding domain, inhibiting microtubule assembly and disrupting mitotic spindle formation, which leads to cell cycle arrest and apoptosis. The combination of these two mechanisms appears to provide enhanced antitumor activity.
