Overview

AMV564 + pembrolizumab is an investigational combination therapy consisting of AMV564, a bivalent bispecific T cell engager targeting CD33 and CD3, and pembrolizumab, a monoclonal antibody immune checkpoint inhibitor targeting programmed cell death protein 1 (PD-1). AMV564 selectively binds to CD33 on myeloid-derived suppressor cells (MDSCs) and CD3 on T cells, depleting immunosuppressive MDSCs and promoting T cell activation and proliferation. Pembrolizumab blocks PD-1, restoring T cell antitumor activity by preventing suppression by its ligand, PD-L1. The combination is being investigated for advanced solid tumors and hematological malignancies, especially where the tumor microenvironment suppresses immune activity through MDSCs and PD-1/PD-L1 pathways. Preliminary clinical studies report good tolerability and evidence of antitumor activity with both agents, alone and in combination[1][5][7].
