Overview
Anakinra + pegsunercept is a combination therapy investigated for the treatment of rheumatoid arthritis. Anakinra is a recombinant form of the human interleukin-1 receptor antagonist (IL-1Ra) that blocks the activity of interleukin-1 alpha and beta by competitively inhibiting their binding to the interleukin-1 type I receptor, thereby reducing inflammation. Pegsunercept is a PEGylated recombinant soluble tumor necrosis factor receptor type I (PEG sTNF-RI), which acts as a decoy receptor to bind and neutralize tumor necrosis factor alpha (TNFα), another key cytokine in inflammatory processes. The combination was studied to assess whether dual inhibition of IL-1 and TNFα would provide additive or synergistic benefit in rheumatoid arthritis; however, clinical trials did not demonstrate added efficacy over monotherapy and showed increased risk of adverse events such as serious infections[2][4][9].
