Overview
Anastrozole + dalpiciclib is an investigational oral combination therapy consisting of two small molecule drugs for the treatment of hormone receptor-positive, HER2-negative advanced or metastatic breast cancer. Anastrozole is a non-steroidal aromatase inhibitor that reduces estrogen production by inhibiting the aromatase enzyme, thereby decreasing tumor growth in estrogen-dependent breast cancers. Dalpiciclib is a selective cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor that blocks cell cycle progression from G1 to S phase by inhibiting phosphorylation of retinoblastoma protein, leading to reduced proliferation of cancer cells. The combination aims to provide synergistic antitumor activity by simultaneously suppressing estrogen-driven signaling and cell cycle progression.
