Drug intelligence / Profile preview
Anbenitamab plus albumin-bound docetaxel is an investigational combination regimen consisting of anbenitamab (KN026), a biparatopic bispecific monoclonal antibody targeting HER2, and HB1801, an albumin-bound formulation of the microtubule-targeting taxane chemotherapy docetaxel. Anbenitamab simultaneously binds to two non-overlapping extracellular domains of HER2 (domains II and IV), which are the binding sites of pertuzumab and trastuzumab, respectively. This dual binding promotes receptor clustering, endocytosis, and lysosomal degradation, thereby inhibiting downstream HER2 signaling. HB1801 is a solvent-free formulation of docetaxel encapsulated in human serum albumin nanoparticles, which avoids the use of polysorbate 80 and ethanol, eliminating the need for steroid premedication and allowing rapid administration at higher concentrations. The combination is being developed by Alphamab Oncology and CSPC Pharmaceutical Group for the treatment of HER2-positive breast cancer, showing significant efficacy in phase 3 trials (such as Neo-Healer and HEALER) in both neoadjuvant and first-line metastatic settings.
