anbenitamab + chemotherapy

Anbenitamab (KN026) + chemotherapy is a combination regimen being evaluated for the treatment of HER2-positive or HER2-expressing cancers. Anbenitamab is a bispecific monoclonal antibody developed by Alphamab Oncology using its proprietary CRIB platform. It simultaneously binds to two non-overlapping epitopes of the HER2 receptor (ECD2 and ECD4), which are the binding sites for pertuzumab and trastuzumab, respectively. This dual binding leads to a more complete blockade of HER2 signaling, enhanced receptor internalization and downregulation, and potent immune-mediated effects such as antibody-dependent cellular cytotoxicity (ADCC). When combined with chemotherapy agents, such as docetaxel or albumin-bound docetaxel, the regimen provides synergistic anti-tumor activity by combining targeted HER2 inhibition with the cytotoxic effects of microtubule disruption. This combination has shown significant clinical activity in Phase II and Phase III trials for first-line HER2-positive recurrent or metastatic breast cancer and gastric/gastroesophageal junction cancers.
