Drug intelligence / Profile preview
JSKN003 + KN026 is an investigational combination therapy being developed by Alphamab Oncology for the treatment of HER2-positive cancers, particularly gastric and gastroesophageal junction (GC/GEJ) cancers. The combination pairs two HER2-targeted biologics: JSKN003 (anbenitamab repodatecan), a biparatopic HER2-targeted antibody-drug conjugate (ADC) carrying a topoisomerase I inhibitor payload, and KN026 (anbenitamab), a recombinant humanized anti-HER2 bispecific antibody that targets extracellular domains II and IV of HER2. By combining the ADC with its parental bispecific antibody, the regimen aims to enhance HER2 receptor clustering, internalization, and bystander killing while blocking HER2 signaling pathways. The combination is being evaluated in a Phase II clinical trial (JSKN003-003, NCT06998771) as a first-line treatment for HER2-positive unresectable locally advanced, metastatic, or resectable gastric cancer, showing promising efficacy and safety.
