Drug intelligence / Profile preview
JSKN003 + capecitabine + SG001 + KN026 is an investigational combination regimen being evaluated for the first-line treatment of HER2-positive advanced or metastatic gastric cancer or gastroesophageal junction (GC/GEJ) adenocarcinoma. The regimen combines two HER2-targeted agents developed by Alphamab Oncology—JSKN003 (anbenitamab repodatecan), a biparatopic HER2-targeting antibody-drug conjugate (ADC) carrying a topoisomerase I inhibitor payload, and KN026 (anbenitamab), a bispecific antibody targeting two distinct HER2 epitopes—with the anti-PD-1 monoclonal antibody enlonstobart (SG001) and the oral chemotherapeutic prodrug capecitabine. By simultaneously targeting multiple HER2 epitopes, delivering a cytotoxic payload, blocking the PD-1/PD-L1 immune checkpoint, and inhibiting DNA synthesis, this multi-pronged regimen aims to overcome resistance and enhance anti-tumor efficacy. In a Phase II clinical trial, this combination (Cohort B) demonstrated a 100% objective response rate (ORR) in patients with first-line HER2-positive GC/GEJ.
