andamertinib + anlotinib

Andamertinib + anlotinib is a combination therapy consisting of a third-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor (TKI) and a multi-targeted receptor tyrosine kinase inhibitor. Andamertinib (ASK120067) is an irreversible inhibitor that selectively targets EGFR sensitizing mutations and the T790M resistance mutation while sparing wild-type EGFR, thereby reducing skin and gastrointestinal toxicities. Anlotinib is a potent small-molecule inhibitor of vascular endothelial growth factor receptors (VEGFR 1-3), fibroblast growth factor receptors (FGFR 1-4), platelet-derived growth factor receptors (PDGFR alpha/beta), and c-Kit. By combining these two agents, the therapy aims to provide dual inhibition of oncogenic signaling pathways and tumor angiogenesis, potentially overcoming resistance to prior EGFR-TKI treatments. This combination is currently being evaluated in clinical trials for patients with locally advanced or metastatic non-small cell lung cancer (NSCLC) who have progressed after treatment with third-generation EGFR-TKIs.
