Drug intelligence / Profile preview
"Androgen deprivation therapy + androgen receptor pathway inhibitor + docetaxel" is a triplet therapy regimen used for the treatment of metastatic hormone-sensitive prostate cancer (mHSPC). The regimen combines three therapeutic components: androgen deprivation therapy (ADT) to lower testosterone levels (typically via LHRH agonists/antagonists or surgical castration); a second-generation androgen receptor pathway inhibitor (ARPI) such as darolutamide, abiraterone acetate, enzalutamide, or apalutamide to block androgen receptor signaling; and docetaxel, a taxane chemotherapy that inhibits microtubule depolymerization. Clinical trials such as ARASENS (evaluating darolutamide) and PEACE-1 (evaluating abiraterone) have demonstrated significant overall survival and progression-free survival benefits with this triplet regimen compared to doublet therapy (ADT plus docetaxel) in patients with biologically aggressive or high-volume mHSPC.
